Abstract: | This image is a 3D reconstruction of a human MRC5 nucleus labelled with cerulean-tagged PCNA. The soluble nuclear material was removed by detergent extraction before the cell was fixed. Confocal Z sections were acquired on a Leica SP5 laser scanning microscope and 3D reconstructions were performed using Amira software. This nucleus measures approximately 18 micrometres across its widest point. Each individual green "blob" inside the nucleus is a replication factory - a site in the cell where the DNA is copied so that the cell can multiply. PCNA is a protein which becomes concentrated in these factories as it is an essential factor required for many of the steps of DNA replication. The nucleus of this cell is very flat as it is a fibroblast, a cell from connective tissue which grows as a flat layer. |
